In the Philippines, vinegar is manufactured by fermenting various starchy, sugary, or alcoholic foodstuffs with Acetobacter, a genus of bacteria named for its tendency to produce acetic acid. Acetic acid, CH3COOH is the compound in vinegar that gives vinegar its sour taste. Vinegar is 4-5% acetic acid by volume. Commonly used feeds in the acetous fermentation manufacture of vinegar include sugar cane, rice wine, pineapple or mango shavings, and coconut juice or sap, etc. The quality and taste are characteristic of the alcoholic liquor from which the condiment is made.
Fraudulent adulteration and or misrepresentation however, has been a problem for commercial vinegar. Solutions of synthetic acetic acid mixed with colorants and flavor enhancers have been marketed as “vinegar”. The prices of these synthetic blends can be so low that they do not allow any price competition between the available authentic vinegars. Fraud may also be in the form of misrepresentation of the fine vinegars, with the cheaper and lower quality products, i.e. substitution of apple cider and balsamic vinegar with cane or other cheaper vinegars.
A procedure based on C-14 analysis of glacial acetic acid isolated from vinegar has been tested at the Philippine Nuclear Research Institute and has been found adequate for the detection of adulteration in vinegar. Radiocarbon assay was done by Low Level Liquid Scintillation Spectrometry. The procedure measures the amount of the radioactive isotope of Carbon, C-14, present in the acetic acid molecules of the samples.
A complementary study focusing on the isotopic ratio, 13C/12C ratio of the same samples was done at the HYDROISOTOP GmbHk Scweitenkirchen, Germany, in an attempt to ascertain the botanical origin, and method of production based on the isotopic signatures of the vinegars. The isotopic ratios of 13C/12C were expressed in terms of permil.
Authentic vinegar samples were acquired, which were prepared by natural fermentation of : sugar cane(VI-131), pineapple juice (VI-135),and mango juice (VI-136). Another type of cane vinegar was prepared by fermentation using an acetator (VI-130).Commercial vinegar were purchased from major supermarkets in the Philippines and were assigned the codes: VI-044,VI-109, VI-116,VI-140,VI-142,VI-152 and VI-158.
On examining the Carbon-14 values gathered from authentic fermented vinegar samples, the range is between 12-15 disintegrations per minute per gram Carbon, dpm/g C --a mean of 14.7 dpm/gC-- was obtained. Potential 100% synthetic samples (VI-0044,VI-0109,VI-0116,VI-0142) showed 14C levels lower than 1 disintegration per minute per gram carbon. Carbon 14 analysis also detected two possible biogenic samples among the commercially available vinegars (VI-0140 & VI-0152) with C-14 values being close to 14.7.
Stable isotope data complements the C-14 data. On examining the delta values obtained from the reference vinegar samples, sample VI-0136 exhibits a signature of a C3 feedstock. C3 plants such as wheat, rye, barley, legumes tubers, and tallow grasses exhibit delta values ranging from (–24)- (-30) per mil. All the other reference authentic samples exhibit signatures characteristic of their C4 feedstock.
Delta values of C4 plants including sugar cane, corn and sorghum are within the (-10) to (-16) per mil range. Possible synthetic/petroleum-derived vinegars exhibit delta values beyond –30 per mil. The acetic acid produced from sugar cane using an acetator presented a fractionation (-10.2 permil.) which allows for it to be differentiated from cane vinegar produced via the conventional fermentation production, not using an acetator.
All the data obtained from 13C/12C measurement corroborate the C-14 data in categorizing between the adulterated and 100 percent natural vinegar samples. In addition, 13C/12C data can further ascertain the plant origin and method of production of the vinegar samples.
Vegetable–derived materials reflect C-14 specific activity characteristic of atmospheric levels at the time the plant was growing. Depletion of these levels from the normal activity of modern Carbon can be used as an indication and measure of the addition of synthetic ingredients such as nonbiogenic acetic acid in vinegar. Synthetic acetic acid produced through the catalytic oxidation of acetaldehyde or from by-products of petroleum distillation can be differentiated from biogenic acetic acid by radiocarbon assay because of a lower C-14 specific activity.
Measurement of natural isotope ratios provide powerful analytical criteria for revealing the botanical origin and method of production, and detection of synthetic acid adulteration in vinegar samples. Acetic acid from different origins exhibit differences in stable isotope composition. Isotope Ratio Mass spectrometry of Carbon isotopes allow discrimination between synthetic and natural products. Isotope ratios can give the botanical origin of prime matters and in certain cases, on the technological process used for food production. The methodology can be applied routinely for the detection of adulteration in vinegar and other fermented products.
